Researchers divided participants into groups receiving either oral orforglipron or placebo. The focus was on weight maintenance over 52 weeks. This design allowed direct comparison of the oral therapy's effects.

Results for Patients Previously on Tirzepatide

Individuals who switched from injectable tirzepatide to oral orforglipron retained 74.7% of their prior weight loss by week 52. In contrast, the placebo group held onto only 49.2% of that loss. This created a treatment difference of about 25.5% in weight reduction.

From week zero to week 52, those on orforglipron experienced an average body weight drop of around 5kg in the tirzepatide cohort. Placebo users regained more weight overall. These figures underscore the pill's role in stabilizing reductions.

Outcomes in Semaglutide Switchers

For participants discontinuing semaglutide injections, orforglipron led to maintenance of 79.3% of earlier weight loss at week 52. The placebo group managed just 37.6% retention. The estimated treatment difference reached 41.7% after one year.

Average weight change from week zero to 52 was about 1kg loss in the orforglipron semaglutide cohort. This pattern showed even stronger benefits compared to the tirzepatide results. Placebo led to greater regain in this subgroup.

Clinical Context and Availability

Researchers observed that patients and some clinicians often believe obesity medications can stop after initial weight loss. In reality, stopping intentional weight loss drugs may cause weight cycling and diminish cardiometabolic gains. Continuous therapy helps preserve benefits.

Oral orforglipron appears as a scalable choice worldwide for holding weight steady post-injectable GLP-1 use. The drug is now available in the US and may launch soon in the UK. These factors support broader access to maintenance options.
