The Regenerative Potential of GHK-Cu in Aesthetic Medicine

GHK-Cu showed a reasonable biological basis for use as a regenerative agent in aesthetic medicine, but the evidence rests mostly on preclinical data and only two RCTs. It enhanced extracellular matrix synthesis, reduced inflammatory markers, and improved wrinkle appearance and patient satisfaction in limited clinical studies. Larger controlled trials with standardized formulations and dosing are needed before consistent therapeutic protocols can be established.

The review included 20 studies: 18 preclinical and 2 RCTs. In preclinical models, GHK-Cu consistently enhanced extracellular matrix synthesis, modulated metalloproteinase activity, promoted angiogenesis and cellular proliferation, and suppressed TGF-beta and IL-6. In clinical studies, GHK-Cu improved patient-reported satisfaction after laser resurfacing and significantly reduced wrinkle volume and depth compared with controls. Microneedle and liposome delivery systems markedly improved transdermal bioavailability and efficacy. The authors concluded that while the biological basis is reasonable, the findings were constrained by methodological variability and a limited number of well-designed clinical trials.

This paper gives GHK-Cu researchers a consolidated review of the peptide's proposed regenerative, anti-inflammatory, and antioxidant mechanisms in aesthetic applications and identifies delivery systems that may improve cutaneous bioavailability. It also documents the small clinical evidence base and methodological variability across studies. It does not establish standardized dosing, formulations, or therapeutic protocols, and it does not prove clinical efficacy beyond the limited RCT evidence.
