Efficacy of tirzepatide in glycemic control and weight management in adults with type 2 diabetes: a systematic review and meta-analysis of real-world studies

Treating adults with type 2 diabetes with tirzepatide in real-world settings was associated with an average HbA1c reduction of 0.91% and weight loss of 9.7 kg across 13 studies. Most users (64%) reached an HbA1c below 7%, and tirzepatide lowered HbA1c and body weight more than control. The evidence is observational and had moderate to serious risk of bias.

Meta-analysis — Systematic review and meta-analysis. Population: Adults with type 2 diabetes in real-world studies. Sample size: 89 296. Follow-up: 3-18 months. Interventions: Tirzepatide. Cited by 1 other paper.

Across 13 real-world studies (N = 89 296; duration 3-18 months) with moderate to serious bias, tirzepatide was associated with a mean HbA1c reduction of 0.91% (95% CI: -1.04 to -0.79), weight loss of 9.7 kg (95% CI: -14.05 to -5.35), and BMI decrease of 2.09 kg/m2 (95% CI: -3.27 to -0.92). Overall, 64% of tirzepatide users reached HbA1c <7%. Tirzepatide lowered HbA1c more than control (MD -0.38%; 95% CI: -0.44 to -0.33) and body weight more than control (MD -6.27 kg; 95% CI: -9.22 to -0.33). Among users, 47%, 23%, 9%, and 4% lost ≥5%, ≥10%, ≥15%, and ≥20% of baseline weight, respectively.

This paper aggregates real-world evidence on tirzepatide for HbA1c lowering and weight management in type 2 diabetes, providing effect sizes and responder rates that researchers can compare with clinical trial data. It does not establish tirzepatide's relative effectiveness against specific diabetes drugs, dose-response relationships, or safety outcomes in routine care.
